Non Natlan characters can still use that set, but you just won't gain the full effect of the passive since they don't have Nightsoul mechanics. You'll only get a 12% dmg buff instead of the full 40%
Depends on which supports but yea. The set works especially well with Natlan characters who are played off field, like Xilonen, Citlali and Ororon

Used 2 Sanctifying Elixir to make myself a new Sands for Chiori and thankfully it turned out to be an upgrade over her previous one.

It wasn't a huge upgrade, but it's an upgrade nonetheless
